Comment The TIAL1 mRNA was found to be enriched in the TIA1 immunoprecipitation relative to control immunoprecipitation (twofold or higher). To determine the functional significance of TIA1 binding to TIAL1 mRNA, the effect of down-regulation of TIA1 on TIAL1 expression was assessed by siRNA knockdown of TIA1. Knockdown of TIA1 expression by its specific siRNA resulted in a significant increase in TIAL1 protein levels.

Comment Biotinylated transcripts corresponding to either the 3â?² untranslated region (UTR) or the coding region (CR) of TIAL1 was incubated with whole-cell lysates. TIA1 associated with the biotinylated TIAL1 transcript and displayed much stronger binding to the 3â?² UTR than the CR fragment.
